چکیده

WHEN it was observed that the majority of SD-identical unrelated individuals stimulated each other in the MLC test, it was postulated in agreement with earlier suggestions by others that MLC activation might be coded by a locus separate from the LA and Four loci." The existence of such a MLC locus is now generally accepted.^About 10% of the MLC tests between SD-identical unrelated persons are negative, indicating that a linkage disequilibrium exists between the MLC locus and the LA and Four loci. It is furthermore likely that SD-identical unrelated individuals who are also LD-identical carry MLC determinants that occur rather frequently.

Genetic and Cellular Aspects of Xenogeneic Mixed Leukocyte Culture Reaction* by Kirsten Fischer Lindahl* and Fritz H. Bach

Xenogeneic lymphocytes can induce proliferative responses in the mixed lymphocyte culture (MLC) ~ which are as strong as those induced by allogeneic lymphocytes (1). It is impossible to determine directly which lymphocyte antigens stimulate the xenogeneic MLC response. The recently described "primed lymphocyte typing" (PLT) assay (2) has made it possible to determine indirectly the genetic cont...

Genetic and cellular aspects of xenogeneic mixed leukocyte culture reaction

The nature of the antigens stimulating xenogeneic lymphocytes was studied using "primed LD typing". Human lymphocytes were sensitized in vitro against mouse spleen cells and restimulated with spleen cells of mouse strains sharing non-H-2 antigens or various regions of H-2 with the initial stimulating strain. The largest thymidine uptake was caused by restimulation with cells from the specific p...

Fine specificity of alloimmune cytotoxic T lymphocytes directed against H-2K. A study with Kb mutants

The fine specificity of alloimmune cytotoxic T lymphocytes (CTL) was investigated in CTL responses across the smallest known H-2 differences, those based on mutation at a single H-2 locus. CTL were generated in all possible mixed lymphocyte culture (MLC) combinations among seven H-2Kb mutants and the mouse strain of origin, C57BL/6 (B6-H-2b). CTL were also generated in all F1 hybrid responder/h...

Mesenchymal stem cells as a reference cell for HLA-typing

Introduction: Recognition of human leukocyte antigens (HLA) is of importance for hematopoietic stem cell transplantation. Any HLA-mismatches between the donor and recipient can cause graft rejection or other complications. In HLA-typing experiments, usage of HLA-known reference cells accompany with HLA-unknown samples is obligatory. Some international centers represent these cells with high e...

I-17: The Ethics of Using IVF and PGD to Conceive A Savior Child

Until the advent of preimplantation genetic diagnosis (PGD) in the 90’s of the last century, the only efficient means to find out whether an offspring has inherited a genetic disease from a couple who are carriers of the defective gene is through prenatal diagnosis (PND). However, if the fetus is found to have the genetic disease, presently the only available treatment is to abort the fetus, an...
